Plate-reader based analysis of extracellular oxygen levels
MitoXpress Xtra assays measure extracellular oxygen consumption by isolated mitochondria, cell populations, small organisms, tissues and enzymes:- ideal for higher through-put analysis.
Easy ‘mix and measure’ plate-based assay using standard 96/384 well plates. Just reconstitute the MitoXpress oxygen probe in water, add to sample, seal with mineral oil, and measure on conventional or time-resolved fluorescent plate readers.
As the test material respires, O2 is depleted in the surrounding environment, which is seen as an increase in probe phosphorescence signal. The assay is reversible: a decrease in oxygen consumption (an increase in O2 levels) is seen as a decrease in probe signal.
Benefits:
- Provides same high quality information as the standard Clarke-electrode, yet is faster, simpler, has a higher throughput, and is more cost-effective
- Fast, easy to run, high-throughput screening of mitochondrial uncouplers and inhibitors
- Allows straight forward generation of IC50s from isolated mitochondrial studies for safety assessment. Screening times are reduced from months to days
- The assay is easily adopted in labs, without the need for specialised equipment.
