The MitoXpress-Xtra assay measures the bacterial oxygen consumption and equates the O2 consumption compound, genetic manipulation or growth conditions. The assay is based on the ability of dissolved O2 to quench the phosphorescence of a soluble, oxygen sensitive probe. As bacteria from the sample grow and respire in test medium, at a certain time point they start to deplete O2 in the surrounding solution, which is seen as an increase in probe signal above baseline level. The time required to reach this threshold signal is then correlated to treatment condition
The assay is a simple ‘mix and measure’ test. Samples are prepared, dispensed in the wells of a standard 96-well plate together with the Redlight oxygen-sensing probe, covered with mineral oil to exclude ambient O2 and measured on a fluorescence plate reader at 30oC in kinetic mode.
