Drug-induced mitochondrial dysfunction can be investigated in vitro using whole cell models, with the advantage that the mitochondrial reticulum is not disrupted by isolation, and biotransformation and transporter activities remain intact.

MitoXpress® Xtra Oxygen Consumption assay enables microplate-based oxygen consumption measurements in cell lines, iPS derived cell models and primary cells, both in 2D and 3D culture, while an additional level of mechanistic information can be generated by parallel monitoring of glycolytic flux using pH-Xtra™ Glycolysis assay.

Specific mitochondrial insult typically results in reduced oxygen consumption and increased glycolytic flux, and so parallel measurements of these key metabolic pathways can increase the capacity to identify specific mitochondrial insult.
